摘要：wehavemeasuredinhumanalveolarcellsthecytoplasmicdistributionofthefluorophorecoumarin-6carriedbysolidlipidnanoparticles(slns)andobservedaperinuclearaccumulationofthefluorescencethatcanbedescribedbyasingleexponentialgrowthalonganideallinejoiningtheplasmamembranetothenuclearborderandbyasigmoidalrelationshipasafunctionoftime.intracellulardistributionwasaffectedbyloweringthetemperaturefrom37to4°candbythedisruptionofcytoskeletonbycytochalasind,butitwasminimallyperturbedbytheinhibitionofatpdependentmolecularmotors.abiophysicalmodelwasdevelopedforanaccumulationofloadedparticlesagainstadiffusiongradientbasedonameanfieldinteractionenergy,whoseoriginweascribetotheactinstructureofthecytoskeleton.theestimatedvaluefortheloaddiffusioncoefficientwasfourandtwoordersofmagnitudelessthanthatoffreecoumarin-6andofslnsinaqueoussolutions,respectively,suggestingthattheloadmoveswithinthecellcytoplasminaformstillreminiscentofthenanocarrierstructure.
